ABTRACT
One of the factors that determine the success of the fertilization process is the ability of sperm to fertilize oocytes. Ejaculated sperm from the male reproductive tract doesn?t have this capability. In the female reproductive tract (cervix) sperm undergo a series of processes that make it has the ability to fertilize an oocyte, called capacitation. Capacitation is an important step for successful fertilization requires the integrity of the plasma membrane and characterized by tyrosine phosphorylation. Definitely, sperm are foreign materials for the female and male body that can stimulate an immune response in the form of antibodies to sperm formation (ASA). This study aims to determine the effect of antisperm antibody from infertile women sera on viability, motility, plasma membrane integrity, as well as the intensity and localization of tyrosine phosphorylation in normal spermatozoa. The design of this study is experimental in vitro. The washed spermatozoa of normal fertile donors incubated with ASA from infertile women sera, with different concentrations (0, 1/1000, 1/100 and 1/10) for 1 hour and 2 hours and then examined the parameters; plasma membrane integrity, intensity and location of tyrosine phosphorylation, viability and motility of spermatozoa. Test results for all measured parameters showed a similar trend, which is incubation of spermatozoa with positive serum ASA with increasing concentration causes a decrease in the percentage of plasma membrane integrity, tyrosine phosphorylation, viability and motility of spermatozoa for 1 hour incubation time, but not so for incubation for 2 hours . But overall all parameters showed a lower percentage value for incubation for 2 hours than 1 hour. Thus, in this study it can be concluded that the ASA from infertile women sera can lower plasma membrane integrity of spermatozoa depending on concentration and incubation time significantly. ASA can also reduce the percentage of tyrosine phosphorylation, viability and motility of spermatozoa depending on concentration and time of incubation, but this decrease was not statistically significant., One of the factors that determine the success of the fertilization process is the ability of sperm to fertilize oocytes.
