Telah dibuat film sambung silang kitosan-tripolifosfat yang mengandung
asiatikosida sebagai pembalut bioaktif untuk luka. Tujuan dari penelitian ini
adalah untuk mempelajari parameter yang berpengaruh dalam pembuatan film
sambung silang kitosan-tripolifosfat, mempelajari karakteristik film yang
dihasilkan, mempelajari profil pelepasan asiatikosida dari film, serta mempelajari
aktivitas penyembuhan lukanya secara in vivo. Film dibuat dalam empat formula
F1, F2, F3, dan F4 dengan memvariasikan konsentrasi tripolifosfat antara lain 0%,
4%, 8%, dan 12% b/b kitosan. Sambung silang kitosan-tripolifosfat dibuat dengan
metode gelasi ionik dan film dibuat dengan metode penguapan pelarut. Cairan
pembentuk film (CPF) dan film yang dihasilkan dikarakterisasi yang meliputi
spektroskopi FTIR, turbidimetri, viskositas, ketebalan, sifat mekanik, daya
mengembang, laju transmisi uap air, kekuatan bioadhesif, profil pelepasan
asiatikosida dari film, dan aktivitas penyembuhan luka secara in vivo pada luka
mekanik terbuka derajat tiga. Hasilnya menunjukkan bahwa film F2, F3, dan F4
memiliki karakteristik yang lebih baik, terutama sifat mekaniknya daripada F1
dengan karakteristik terbaik ditunjukkan oleh F4. Persen kumulatif pelepasan
asiatikosida pada jam ke enam dari film F1, F2, F3, dan F4 berturut-turut 84,8%,
72,1%, 73,4%, dan 72,0% dengan kinetika pelepasan dikontrol oleh proses difusi
dan erosi. Film sambung silang kitosan-tripolifosfat (F4) belum menunjukkan
aktivitas penyembuhan luka yang lebih baik dibandingkan kontrol dan aktivitas
yang ditunjukkan tidak berbeda secara bermakna (p > 0,05) pada jenis luka yang
diujikan (luka kering).

<b>ABSTRACT</b><br>
Cross-linked chitosan-tripolifosfat films containing asiaticoside have been
prepared as bioactive dressing. The objectives of this research were to study the
parameters that affect in preparation of cross-linked chitosan-tripolyphosphate
films, to study the characteristics of the resulting films, to study the release profile
of asiaticoside from the films, and to study in vivo wound healing activity. The
Films were formulated in four formulas termed F1, F2, F3, and F4 by varying the
concentration of tripolyphosphate including 0%, 4%, 8%, and 12% w/w of
chitosan. Cross-linked chitosan-tripolyphosphate was prepared by ionic gelation
technique and the films were prepared by casting/ solvent evaporation technique.
Film-forming fluids (CPF) and the resulting films were characterized, including
spectroscopy FTIR, turbidimetry, viscosity, film thickness, mechanical properties,
swelling degree, water vapor transmission rate, bioadhesive property, release
profil of asiaticoside from the film, and in vivo wound healing activity on third
degree mechanical open wound. The result showed that F2, F3, and F4 films had
better characteristics especially in mechanical properties than F1 film and the best
characteristics was showed by F4 film. Cumulative release of asiaticoside at sixth
hours from F1, F2, F3, and F4 films respectively were 84,8%, 72,1%, 73,4%, and
72,0% with the release kinetics were controlled by diffusion and erosion process.
Chross-linked chitosan-tripolyphosphate film (F4) has not showed better wound
healing activity than control and the activity wasn't significantly different on the
