Cedera ginjal akut (acute kidney injury/AKI) merupakan salah satu penyakit ginjal dengan tingkat morbiditas dan mortalitas yang tinggi pada manusia. Salah satu kondisi yang tergolong dalam AKI adalah ischaemia-reperfusion injury (IRI), ditandai dengan terjadinya iskemia dan diikuti dengan reperfusi. Penetapan hewan model IRI diperlukan sebagai suatu metodologi untuk mensimulasikan perubahan patofisiologi yang terjadi dan mengamati tercapainya kondisi klinis IRI yang paling representatif. Penelitian ini membahas metode bedah bilateral renal-pedicle clamping untuk menginduksi terjadinya iskemia pada hewan model IRI dengan galur tikus Sprague-Dawley. Sebanyak 24 ekor hewan uji dikelompokkan menjadi dua berdasarkan waktu reperfusinya terlebih dahulu, yaitu 24 jam dan 14 hari. Kedua kelompok tersebut masing-masing dibagi kembali menjadi empat kelompok (n=3). Kelompok normal tidak diberi perlakuan bilateral renal-pedicle clamping, namun kelompok I, II, dan III diberi perlakuan tersebut dengan durasi clamping 15, 30, dan 45 menit secara berurutan. Selanjutnya, dilakukan analisis terhadap perubahan parameter struktural ginjal dengan pengamatan histopatologi menggunakan pewarnaan PAS (periodic acid-Schiff). Parameter yang dinilai adalah tingkat keparahan cedera tubular yang terjadi. Hewan uji dari kelompok I, II, dan III menunjukkan membaiknya kondisi cedera pada waktu reperfusi 14 hari dari kondisinya pada waktu reperfusi 24 jam, dengan kelompok II yang menunjukkan perbedaan paling signifikan. Dapat ditarik kesimpulan bahwa durasi clamping selama 30 menit menyebabkan tercapainya kondisi klinis IRI yang paling representatif dan menunjukkan pemulihan kondisi cedera yang paling signifikan dalam jangka waktu reperfusi 14 hari pada hewan model IRI.

<i><b>ABSTRACT</b>
Acute kidney injury (AKI) is a kidney disease with high levels of morbidity and mortality in humans. One condition that is classified as AKI is ischaemia-reperfusion injury (IRI), characterized by the occurrence of ischaemia and followed by reperfusion. Establishment of IRI animal models is needed as a methodology to simulate pathophysiological changes that occur and observe the achievement of IRI's most representative clinical conditions. This study discusses bilateral renal-pedicle clamping surgical method to induce ischaemia in IRI model animals with the Sprague-Dawley mouse strain. A total of 24 animals were grouped into two based on their reperfusion time, 24 hours and 14 days. The two groups are each subdivided into four groups (n=3). Normal groups were not treated with bilateral renal-pedicle clamping but group I, II, and III were given the treatment with clamping duration of 15, 30, and 45 minutes respectively. Then, analysis of renal structural parameters changes was performed with histopathological observation using PAS (periodic acid-Schiff) staining. Parameter to be assessed is the severity of tubular injury. Animals from group I, II, and III showed an improvement in injury condition at the reperfusion time of 14 days from their condition at reperfusion time of 24 hours, with group II showing the most significant difference. It can be concluded that the clamping duration of 30 minutes leads to the achievement of the most representative clinical IRI conditions and shows the most significant recovery of injury conditions within the 14-day reperfusion period in IRI animal models.<i/></description><identifier>http://lontar.ui.ac.id/detail?id=20504355</identifier><recordID>20504355</recordID></dc>
