Maka itu, kanker serviks membutuhkan tata laksana yang adekuat untuk menurunkan mortalitas pasien. Tata laksana yang saat ini kerap dijalankan memiliki beragam efek samping dan belum dapat menjamin kesembuhan pasien, sehingga dibutuhkan alternatif lain sebagai pendukung yang minim efek samping, misalnya dengan penggunaan herbal. Salah satu tanaman yang berpotensi dikembangkan sebagai obat herbal adalah tanaman kunto dewo (Kigelia pinnata) yang kerap digunakan secara tradisional untuk mengobati berbagai gejala dan penyakit, serta memiliki efek antiinflamasi, antibakterial, antioksidan, hingga antikanker. Tujuan: Mengetahui kandungan senyawa fitokimia, total flavonoid, total fenol, dan aktivitas antikanker in vitro ekstrak etanol kulit dan daging buah kunto dewo (Kigelia pinnata) terhadap sel HeLa. Metode: Kulit dan daging buah Kigelia pinnata diperoleh dari daerah sekitar candi Borobudur, Magelang, Jawa Tengah. Kulit dan daging buah Kigelia pinnata dikeringkan, lalu masing-masing dimaserasi dengan pelarut etanol, selanjutnya filtrat hasil penyaringan diuapkan hingga menghasilkan ekstrak etanol kulit dan daging buah Kigelia pinnata. Pada kedua ekstrak dilakukan analisis fitokimia melalui penapisan fitokimia, analisis kromatografi lapis tipis (KLT), dan penentuan total flavonoid dan total fenol. Ekstrak kemudian diuji aktivitas antikankernya secara in vitro terhadap sel HeLa dengan metode MTT. Hasil: ekstrak etanol kulit dan daging buah Kigelia pinnata mengandung senyawa tanin, flavonoid, steroid, dan triterpenoid. Analisis KLT menunjukkan bahwa ekstrak etanol Kigelia pinnata mengandung 4 komponen senyawa fitokimia. Ekstrak etanol kulit buah Kigelia pinnata (EEKB) memiliki kadar total fenol dan flavonoid masing-masing sebesar 9,02 mg ekuivalen asam galat(GAE)/g dan 6,3 mg ekuivalen kuersetin(QE)/g sedangkan ekstrak etanol daging buah Kigelia pinnata (EEDB) memiliki kadar total fenol dan flavonoid masing-masing sebesar 12,96 mgGAE/g dan 0,91 mgQE/g. Kedua ekstrak menunjukkan aktivitas antikanker in vitro yang moderat terhadap sel HeLa, dengan nilai IC50 masingmasing sebesar 127,02 µg/mL untuk EEKB dan 92,13 µg/mL untuk EEDB. Therefore, cervical cancer requires adequate management to reduce patient mortality. The current management often has a variety of side effects and cannot guarantee the patient's recovery, so that other alternatives are needed to support minimal side effects, for example by using herbs. One of the plants that have the potential to be developed as herbal medicine is the kunto dewo (Kigelia pinnata) which is often used traditionally to treat various symptoms and diseases, and has anti-inflammatory, antibacterial, antioxidant, and anti-cancer effects. Aim: To determine the content of phytochemical compounds, total flavonoids, total phenols, and in vitro anticancer activity of the ethanol extract of the skin and flesh of kunto dewo (Kigelia pinnata) against HeLa cells. Method:. The skin and flesh of Kigelia pinnata fruit were obtained from the area around the Borobudur temple, Magelang, Central Java. The skin and flesh of the Kigelia pinnata fruit were dried, then each macerated with ethanol solvent, then the filtered filtrate was evaporated to produce an ethanol extract of the skin and flesh of the Kigelia pinnata fruit. In both extracts, the phytochemical analysis was carried out through phytochemical screening, thin layer chromatography (TLC) analysis, and determination of total flavonoids and total phenols. The extract was then tested for its anticancer activity in vitro against HeLa cells using the MTT method. Result: The ethanol extract of Kigelia pinnata fruit skin and flesh contained tannins, flavonoids, steroids, and triterpenoids. TLC analysis showed that the ethanol extract of Kigelia pinnata contained 4 phytochemical compounds. The ethanol extract of Kigelia pinnata fruit peel (EEKB) has a total phenol and flavonoid content of 9.02 mg gallic acid equivalent (GAE) / g and 6.3 mg quercetin (QE) / g, while the ethanol extract of Kigelia pinnata pulp (EEDB) has total phenol and flavonoid levels of 12.96 mgGAE / g and 0.91 mgQE / g, respectively. Both extracts showed moderate in vitro anticancer activity against HeLa cells, with IC50 values of 127.02 µg / mL for EEKB and 92.13 µg / mL for EEDB, respectively.
