Mesenchymal stem cell-conditioned medium (MSC-CM) known as secretome, contains a lot of regenerative molecules known as cytokines and growth factors. The objective of this study was to investigate the anti-aging effect of MSC-CM in mice skin model induced by D-galactose. Thirty-nine of 6 weeks old mice were divided into 4 group, control group, D-galactose (D-gal) group, saline treated group and MSC-CM treated group. D-galactose were injected once a week for 8 weeks to induce skin aging at the dose rate of 1000 mg/kg BW. After skin aging induction, mice were injected subcutaneously with saline in saline treated group and MSC-CM in MSC-CM treated group once a week. Mice were sacrificed before D-gal induction, after D-gal induction and after treated by saline and MSC-CM. Skin tissues were collected and processed by paraffin method. Sample tissues were stained with Hematoxyline-Eosin and Mallory Aniline Blue staining method. MSC-CM recovered the skin structure and morphology as indicated by the enhance of epidermal and dermal thickness, enhance vascularization and hair follicle formation, and very high density of collagen fibers deposition.
