- The present study was carried out to evaluate swelling degree and dissolution profile of famotidine from matrix polyelectrolyte complexes (PEC) chitosan and xanthan gum that prospective for gastroretentive system. PEC was made with chitosan solution and xanthan gum solution with ratio 1:4. The Matrix was made by direct compressed method in 6 formulas, which were based on PEC concentration, 40%, 60%, 80% and 92%, which compared with two formula as the control, 92% matrix of chitosan and xanthan gum. The swelling degree was determined in hydrochloric acid buffer medium pH 1.2 for 3 hours. The result showed that increasing PEC in the tablet matrix causing the swelling degree increased. The dissolution test was carried out by using dissolution testing apparatus 2 (paddle method) in a buffer medium pH 1.2 hydrochloric acid at temperature 37 ± 0.5 0C, speed 50 rpm for 8 hours. The results showed a tendency to follow zero-order kinetics for all formulas, and the release profiles of famotidin was controlled by diffusion mechanism. Formula D (PEC 40%) showed the best dissolution profile. Keyword : chitosan, xanthan gum, famotidine, polyelectrolyte complexes.
